Changes of MiR-1281 and MiR-122 in Peripheral Blood of DR Patients and Their Relationship with the Severity of Retinopathy

Abstract:  Objective To investigate the changes and clinical significance of miR-1281 and miR-122 in peripheral blood of patients with diabetic retinopathy DR. Methods 55 patients with type 2 diabetes without retinopathy NDR group), 42 patients with non-proliferative DR NPDR group), 30 patients with proliferative DR PDR group), and 40 healthy people control group were selected as the study subjects. The relative expression of miR-122 and miR-1281 in serum was determined by real-time fluorescence quantitative polymerase chain reaction. Results The serum miR-122 level in NPDR group and PDR group was significantly higher than that in control group and NDR group and that in PDR group was significantly higher than that in NPDR group P<0.05. The level of miR-1281 in NDR group NPDR group and PDR group was significantly higher than that in the control group and the expression level of miR-1281 in NDR group NPDR group and PDR group was significantly higher than that in the control groupP<0.05. Serum miR-122 and miR-1281 were significantly and positively correlated with the course of disease glycosylated hemoglobin and HOMA-IR index. The area under the curve of miR-122 and miR-1281 for differentiating proliferative DR was 0.81 and 0.86 respectively. Conclusion The expression of serum miR-122 and miR-1281 increased in DR patients which can be used as an index to evaluate the severity of DR.
