摘要:

目的:探讨组蛋白甲基转移酶EZH2在神经细胞发育中的作用。方法:建立小鼠胚胎干细胞体外诱导分化成神经细胞
的体系,通过观察细胞形态和实时定量RT-RCR验证诱导分化效果,并通过Western blot方法检测EZH2蛋白在胚胎干细胞分
化过程中的表达变化。结果: 在全反式维甲酸(RA)诱导的E14小鼠胚胎干细胞分化为神经细胞的过程中,观察到EZH2蛋白
在胚胎干细胞状态以及诱导分化的第1天表达量比较高,第2天开始表达量逐渐降低,到第5天实验结束,降至最低。结论:观
察到EZH2蛋白在神经细胞发育过程中的特异性表达变化,为下一步研究EZH2的作用机制打下了基础。

Abstract:

Objective: To study the role of histone methyltransferase EZH2 in the neuron development. Methods: Mouse embryonic
stem cells were induced into neural cells by all-trans retinoic acid(RA)in vitro, which was verified by cell morphology and real-time
quantitative RT- PCR. EZH2 protein expression level during the differentiation procedure was evaluated by Western blot assays.
Results: In 5 days of E14 embryonic stem cells differentiation, the protein expression levels of EZH2 were the highest in the state of
embryonic stem cells and 1d after induction, the protein expression levels then declined rapidly. At the end of the 5d, protein expression
level was the lowest. Conclusion: This research observed the specific expression change of EZH2 protein in the differentiation of neuron,
which is the foundation for further study of EZH2.
