Study on Anti-Oxidative Stress Effect of Xigui Crude Polysaccharide for Hypoxic Mice

Abstract:

 Objective: To study anti-oxidative stress effect of Xigui crude polysaccharide for hypoxic mice. Methods: Kunming mice were randomly divided into normal group, hypoxic model group, salidroside group(360 mg/kg), Xigui crude polysaccharide low dose group(100 mg/kg), medium dose group (200 mg/kg) and high dose group (400 mg/kg). The survival time of the first batch of mice put in hypoxic environment was measured. The superoxide dismutase(SOD) activity, glutathione peroxidase Px(GSH-Px) activity, malondialdehyde (MDA) content, acetylcholinesterase (AChE) activities in liver tissue and serum, and serum nitric oxide (NO) content in the second batch of mice were measured. Results: Each dose group of Xiguei crude polysaccharide could prolong the survival time of mice, significantly decrease the content of MDA in serum(P<0.01), and up-regulate the activities of SOD and GSH-Px in serum, and the high dose group had the best effect(P<0.05). The medium and high dose groups of Xiguo crude polysaccharide could significantly decrease the content of NO and the activity of AChE in serum of mice(P<0.05), significantly increase the activity of SOD in liver tissue of mice(P<0.05), decrease the content of MDA in liver tissue of mice and increase the activity of GSH-Px in liver tissue of mice, and the high dose group had the best effect (P<0.05). The AChE activity in liver tissue of mice could be decreased in the middle dose group of Xiguei crude polysaccharide(P<0.05). Conclusion: Xigui crude polysaccharide can effectively prolong the survival time of mice under hypoxia and has a better anti-oxidative stress effect.
