Effectiveness Evaluation of an Improved Method for Soil Nematodes Isolation

Abstract:

Objective: To evaluate the further improved version(improved method 2) of improved Baermann funnel method (improved method 1) reported in the literature. Methods: Soil artificially added with quantitative nematodes and natural farmland soil were used as experimental materials, and then the isolation rate of soil nematodes and costs of shallow dish method, Baermann funnel method, improved method 1, improved method 2 and LUDOX TM suspension method were measured. Results: The isolation rate of nematodes by the improved method 2 showed no significant difference from that by LUDOX TM suspension method, but is significantly higher than those by other methods(P<0.01). Conclusion: Improved method 2 has obvious advantages in terms of nematode isolation rate, operation time and cost.
