Determination of Content of Total Flavones from the Plant of Arctium lappa L.

摘要:

目的:测定牛蒡植株不同部位总黄酮的含量。方法:采用L9(34)正交设计试验,对提取时间、料液比、乙醇浓度、提取次数4因素进行考察,以确定最佳提取工艺;以芦丁为对照,用亚硝酸钠-硝酸铝-氢氧化钠比色法测定总黄酮含量。结果:最佳提取工艺为:乙醇浓度55%,料液比1颐70,提取时间1.5 h,提取次数2次;芦丁在4.00~100.00 滋g/mL范围内线性关系良好,r=0.999 8,总黄酮平均回收率为97.7%,RSD为2.5%(n=6)。结论:该提取工艺稳定可行,含量测定方法准确。

Abstract:

Objective: To determine the content of the total flavones from the Arctium lappa L.. Methods: The best extraction
technology was determined by orthogonal experiment design, including extraction time,solid-liquid ratio,ethanol concentration and the extraction times. The content of total flavones was determined by absorption spectrometry with rutosids as reference substance.Results: Optimum extraction technology was as follows: 55% ethanol,ratio of material to liquid 1:70,time 1.5 h,two times. The calibration curve had good linear relationship in the range of 4.00~100.00 μg/mL. The average recovery was 97.7%,RSD=2.5%(n=6).Conclusion: The extraction process is stable and feasible. The quantitative method is accurate.
