Effectiveness Evaluation of an Improved Method for Soil Nematodes Isolation

摘要: 目的:对文献报道的贝尔曼漏斗改良法(改良法1)进一步改良(改良法2),并进行评估。方法:以人工添加定量线虫土壤和自然土壤为实验材料,分别测定浅盘法、贝尔曼漏斗法、改良法1、改良法2和LUDOX TM悬浮法对土壤线虫的分离率,并分析不同方法的实验成本。结果:改良法2与LUDOX TM悬浮法的线虫分离率无显著差异,但显著高于其他方法(P< 0.01)。结论:改良法2在线虫分离率、操作耗时与成本方面具备更多优势。

Abstract:

Objective To evaluate the further improved versionimproved method 2 of improved Baermann funnel method improved method 1 reported in the literature. Methods Soil artificially added with quantitative nematodes and natural farmland soil were used as experimental materials and then the isolation rate of soil nematodes and costs of shallow dish method Baermann funnel method improved method 1 improved method 2 and LUDOX TM suspension method were measured. Results The isolation rate of nematodes by the improved method 2 showed no significant difference from that by LUDOX TM suspension method but is significantly higher than those by other methodsP<0.01. Conclusion Improved method 2 has obvious advantages in terms of nematode isolation rate operation time and cost.
