摘要: 目的:探讨砷及其代谢物对人肺癌细胞A549cIAP1cIAP2XIAP mRNA表达的影响。方法:以不同浓度亚砷酸钠(NaAsO2)和60  μmol/L 甲基胂酸(MMA)、二甲次胂酸(DMA)染毒A549细胞48 h,用CCK-8试剂对染毒后的细胞进行细胞活力检测。基于细胞活力结果,用不同浓度NaAsO260  μmol/L MMADMAA549细胞染毒,采用Caspase-3活性试剂检测染毒后细胞凋亡和坏死情况,并通过实时荧光定量聚合酶链式反应(PCR)对染毒后细胞cIAP1cIAP2XIAP mRNA表达水平检测。结果:随着砷浓度的增加,细胞活力下降;NaAsO2染毒组随剂量的增加,细胞凋亡与坏死增加,MMADMA也可诱导少量细胞凋亡。实时荧光定量PCR结果显示,NaAsO2染毒组及其代谢物MMAcIAP1cIAP2XIAP mRNA表达与对照组(0 μmol/L NaAsO2)相比明显降低(P<0.05);但DMA对这3种基因mRNA表达无显著影响。结论:NaAsO2及其代谢产物MMA可显著降低A549细胞中cIAP1cIAP2XIAP mRNA的表达,但其代谢产物DMA对这3种基因mRNA的表达无显著影响。

Abstract:

Objective To investigate the effects of arsenic and its metabolites on the expression of cIAP1cIAP2 and XIAP in  human lung aocarcinoma cell A549. Methods A549 cells were exposed to different concentrations sodium arsenite NaAsO2), 60 ?mol/L monomethylarsonic acid MMA and dimethylarsinic acid DMA for 48 h and the cell viability was detected with CCK-8 reagent. Based on the results of cell viability A549 cells were poisoned with different concentrations of NaAsO2 60 ?mol/L MMA and DMA and Caspase-3 active reagent was used to detect the apoptosis and necrosis of cells after being poisoned. The mRNA expression levels of cIAP1 cIAP2 and XIAP were detected by real-time fluorescent quantitative polymerase chain reactionPCR. Results With the increase of arsenic concentration cell viability decreased. With the increase of dose apoptosis and necrosis increased in NaAsO2 group meanwhile MMA and DMA could also induce a small number of cell apoptosis. The results of real-time fluorescent quantitative PCR showed that the expression of cIAP1 cIAP2 and XIAP mRNA in the NaAsO2 exposure group and its metabolite MMA was significantly lower than that in the control groupP<0.05); but DMA had no significant effect on the mRNA expression of these three genes. ConclusionNaAsO2 and its metabolite MMA can reduce the expression of cIAP1 cIAP2 and XIAP mRNA in A549 cells but its metabolite DMA had no significant effect on the mRNA expression of these three genes.
