Abstract:

Objective: To investigate the effect of HDAC1siRNA on phenotypic changes and function of TGF-β1-induced type Ⅱ
alveolar epithelial cells(RLE-6TN)to EMT. Methods: RLE-6TN cells were cultured and treated with HDAC1 siRNA, then TGF-β1
was added to the cells before collecting cells. The morphological changes were examined by phase-contrast microscope while the
phenotypic changes were examined by the immune fluorescence method. The protein and mRNA expression of HDAC1, HDAC8, Ecad
and α-SMA of RLE-6TN Cells were assayed by Western Blot and Real-time RT-PCR, respectively. The migration in vitro of cells
was assayed by Transwell chambers. Results: Morphologically, TGF-β1 could induce the EMT process of the RLE-6TN cells; after
transfection, cells were polygonal and like the flagstones. Under fluorescence microscope, the cells had expression of α-SMA with
increasing expression of HDAC1 in the group treated with TGF-β1, transfection group is the opposite. At the mRNA level, TGF-β1
could down-regulate the expression of E-cad with up-regulation the HDAC1 and HDAC8 expressions; after transfection, the HDAC1
and HDAC8 expressions were down-regulated with up-regulation the α-SMA expression. At the protein level, TGF-β1 could induce
the down-regulation expression of E-cad with up-regulation expression of the α-SMA and HDAC1, transfection group is the opposite.
In vitro migration ability, TGF-β1 could increase migration cells while transfection group reduced. Conclusion: HDAC1 siRNA could
partially reverse the EMT of RLE-6TN cells induced by TGF-β1 in vitro.
