Inhibition of K562 Cell Proliferation by p53 Protein Regulated by Periplaneta americana Extract C-3

Abstract: Objective To investigate the effect of Periplaneta americana extract C-3 on the proliferation of human leukemia cell K562. Methods K562 cells were treated with different concentrations of Periplaneta americana extract C-3 and the effects of C-3 on the proliferation of K562 cells were detected by CCK-8 assay. Colony formation assay was used to detect its effect on proliferation of K562 cells. The cell cycle of K562 cells was detected by flow cytometry. The expression of phosphorylated p53 protein was detected by Western blot. ResultsPeriplaneta americana extract C-3 could inhibit the proliferation of K562 cells and the optimal concentration and duration of action were 80 μg/mL and 72 h respectively. Periplaneta americana extract C-3 reduced the colony formation rate of K562 cells. The proportion of K562 cells blocked in G0/G1 phase increased while that in S phase decreased. The expression of phosphorylated p53 protein was up-regulated. Conclusion Periplaneta americana extract C-3 inhibits the proliferation of K562 cells by regulating p53 protein.
