Research on the Point Mutation of K-ras Gene in Human Malignant Pleural Mesothelioma

Abstract:

ObjectiveTo investigate K-ras gene point mutation in malignant pleural mesotheliomaMPMtissue of patients from the crocidolite-contaminated area in Dayao ountyYunnan rovince. MethodsPolymerase chain reactionPCRand single-strand conformation polymorphism and PCR-DNA sequencing were used to detect the point mutations of K-ras codons 1213 and 61 in pleural mesothelium of 59 MPM patients and 13 non-MPM patients. ResultsThere was no point mutation of K-ras gene in pleural mesothelial tissues of 59 MPM patients and 13 non-MPM patientsand the detection rate of K-ras gene point mutation was 0.0 %. ConclusionThe proportion of K-ras gene mutation type in MPM tissue is relatively lowwhich suggests that K-ras proto-oncogene point mutation does not play a key role in the induction of MPM in the crocidolite-contaminated area of Dayao countyYunnan Province.
