在校大学生UCP1基因-3826 A>G位点多态性与体质指数的相关性分析

Correlation Analysis of UCP1 Gene -3826 A > G Locus Polymorphism and Body Mass Index in College Students

摘要:

[摘要] 目的:探讨在校大学生UCP1基因-3826 A>G位点多态性与体质指数(BMI)之间的关系。方法:随机选取308例在校大学生,从口腔黏膜脱落细胞中提取基因组DNA,记录受试者身高、体质量,利用KASP技术检测UCP1基因-3826 A>G位点多态性,并对其基因型频率、等位基因频率及不同基因型与BMI的关系进行统计分析。结果:308例受试者中男性96例(占31.2%),女性212例(占68.8%),AA基因型(野生型)64例(占20.8%),AG基因型(杂合变异型)216例(占70.1%),GG基因型(纯合变异型)28例(占9.1%),不同性别受试者各基因型构成比差异无统计学意义。UCP1基因-3826 A>G位点的突变频率随着BMI的增加而增高,差异有统计学意义(P<0.05);风险评估显示,与体质量正常组相比,GG基因型会使体质量增加的风险增高3.875倍。结论:UCP1基因-3826 A>G位点多态性与BMI之间存在一定的关联性,该位点发生纯合变异会使体质量增加的风险提高。

Abstract:

AbstractObjectiveTo explore the relationship between the UCP1 gene -3826 A>G locus polymorphism and body mass index BMIin college students. Methods308 college students were randomly selectedand the genomic DNA were extracted from exfoliated cells of oral mucosa. The height and body weight of the subjects were recorded. The polymorphism of UCP1 gene -3826 A >G locus was detected by KASP technology. The genotype frequenciesallele frequencies and the relationship between different genotypes and BMI were statistically analyzed. ResultsAmong the 308 subjectsincluding 96 cases31.2%were male and 212 cases68.8%were female64 cases 20.8%)  were AA genotype wild type), 216 cases 70.1%were AG genotype heterozygous variant), and 28 cases9.1%were GG genotypehomozygous variant. There was no statistical significance in the composition ratio of genotypes between different genders. The mutation frequency of UCP1 gene -3836 A>G locus increased with the increase of BMIand the difference was statistically significant P<0.05); the risk assessment showed that the GG genotype increased the risk of body weight increase by 3.875 times compared with the normal group. ConclusionThe results indicated that the UCP1 gene-3826 A>G locus polymorphism was associated with the BMIthe homozygous variant at this locus will increase the risk of body weight increase.
