齿瓣石斛水提物对环磷酰胺免疫抑制模型小鼠的影响

Effects of Dendrobium devonianum Water Extract on Cyclophosphamide Immunosuppressive Model Mice

摘要: 目的:探讨齿瓣石斛水提取物(DDWE)对环磷酰胺(CTX)诱导的免疫抑制模型小鼠的影响。方法:将CTX免疫抑制模型小鼠随机分组,连续灌胃给药30 d后,检测小鼠腹腔巨噬细胞吞噬功能、脾淋巴细胞增殖能力、脾脏指数、T淋巴细胞亚群与外周血细胞数量;酶联免疫吸附试验(ELISA)检测血清中免疫球蛋白G IgG)、免疫球蛋白A IgA)、免疫球蛋白M IgM)、肿瘤坏死因子-α TNF-α)与白细胞介素-2 IL-2)的水平。结果:与模型组相比,DDWE各剂量组不同程度加强巨噬细胞吞噬功能与淋巴细胞增殖能力,剂量依赖性提高脾指数、白细胞数、CD4+/CD8+水平以及血清中IgGIgAIgMTNF-α与IL-2水平。结论:DDWE通过加强免疫细胞活性,促进免疫细胞因子的产生与释放,起到增强CTX免疫抑制小鼠免疫功能的作用。

Abstract:

Objective To investigate the effects of Dendrobium devonianum water extract(DDWE) on immunosuppressive model mice induced by cyclophosphamide CTX. Methods CTX immunosuppressive model mice were randomly divided into groups. After 30 days of continuous intragastric administration the phagocytic function of mice peritoneal macrophages the proliferation ability of mice splenic lymphocytes splenic index T lymphocyte subsets and the number of peripheral blood cells were measured the level serum of immunoglobulin GIgG), immunoglobulin AIgA), immunoglobulin MIgM), tumor necrosis factor-α(TNF-α) and interleukin-2IL-2 were detected by ELISA method.  Results Compared with the model group each dosage group of DDWE enhanced the phagocytic activity of macrophages and proliferation ability of lymphocytes to varying degrees. And a dose-dependent manner increased the splenic index the number of white blood cells the level of CD4+/CD8+ and the levels of serum of IgG IgA IgM TNF-α, and IL-2. Conclusion DDWE strengthened the immune function of CTX immunosuppressive mice by improving the activity of immune cells and promoting the production and release of immune cytokines.
