Determination of Shikimie Acid in Pinus armandi by HPLC

Abstract:

Objective: To establish an analytical method to respectively investigate shikimic acid in different parts of Pinus armandi based on high performance liquid chromatographic experiments. Methods: Agilent Zorbax XDB-C18 column (4.6 mm×150 mm,5μm) was used to develop the new analytical method while the mobile phase was methanol and 1% phosphoric acid solution(3:97);the flow rate was 0.8 mL.min-1, and the detection wavelength was 214 nm; the column temperature was set at 25°C. Results: The linear relationship between the peak area and content of shikimic acid was observed in the range of 0.041 5-0.581 0μg(r=0.999 9);the average recovery was 98.74%, and RSD was 0.74% (n=9). All parts of P. armandi contained shikimic acid, but the contents significantly varied from part to part. Conclusion: P. armandi can be a new resource of shikimic acid for exploitation and utilization.
