Determination of Chlorogenic Acid in Alnus ferdinandi-coburgii by HPLC

Abstract:

Objective: To establish an HPLC method for the determination of chlorogenic acid in Alnus ferdinandi -coburgii.
Methods: We applied highly effective liquid phase chromatography, with Venusil X BP-C18 column (150 mm×4.6 mm,5 μm)and
mobile phase consisted of acetonitrile-0.4% phosphoric acid solution(8:92)at a flow rate of 1.0 mL/min. The detection wavelength was
set at 324 nm, and column temperature was 25 0C. Results: There was good linearity in the range of 0.108 8-1.088 μg (r=0.999 9)
of chlorogenic acid. The average recovery was 98.98%, RSD was 0.77%(n=6). Conclusion: The method above was simple, sensitive,
accurate, reproducible, and could be applied in the measurement of chlorogenic acid in Alnus ferdinandi-coburgii.
