Abstract:

Objective: To study the effects of grape seed proanthocyanidin extract(GSPE)on antioxidation of hepatic ischemia
reperfusion injury in rats, and the expression of apoptosis- related proteins Bcl- 2 and Bax. Methods: Thirty SD male rats were randomly divided into three groups with 10 rats each .i.e. sham operation group, HIRI model group and GSPE preconditioning group (160 mg/kg). The content of GSH-Px, SOD, and MDA in hepatic tissues, and the content of ALT and AST in serum were assayed 3 hours later after reperfusion. The morphological variations of hepatic tissues in each group were compared by means of optical microscope. Effects of GSPE on Bcl-2 and Bax in hepatic tissues were tested by means of immunohistochemisty. Results: The content of ALT and AST in serum and MDA in hepatic tissues decreased significantly(P<0.05), and the content of GSH-Px and SOD in hepatic tissues increased remarkably(P<0.05)in GSPE preconditioning group, compared with HIRI model; Observed by the optical
microscope, the degree of liver cell injury in GSPE preconditioning group was remarkably slighter than that of HIRI model group. Immunohistochemical results showed that the content of Bcl-2 increased and Bax decreased significantly in hepatic tissues, compared with that in HIRI model(P<0.05). Conclusion: GSPE had some effects on antioxidation and anti-apoptosis of hepatic ischemia reperfusion injury.
