Effect of Scutellarin on CYP2C9 in Chang Liver Cells

Abstract: 〔Abstract〕Objective: To study the effect of scutellarin on the expression of CYP2C9 in Chang Liver cells. Methods: The effect of
scutellarin on proliferative activity of Chang Liver cells was detected by MTT assay. The relative expression of CYP2C9 mRNA and
protein was detected by RT- PCR and Western- blot respectively. Results: MTT results showed that scutellarin inhibited the
proliferation of Chang Liver cells in a concentration-dependent manner, and the half inhibitory rate(IC50)was 1.87 mmol/L. RT-PCR
results showed that the expression of CYP2C9 mRNA significantly decreased in 5.0, 10.0, 20.0, 40.0 μmol/L scutellarin groups(P<
0.05), compared with DMSO group. Western- blot results showed that the expression of CYP2C9 protein in 20.0, 40.0 μmol/L
scutellarin groups significantly decreased(P<0.05), compared with DMSO group. Conclusion: Scutellarin has different degree of
inhibitory action on the expression of CYP2C9 mRNA and protein in Chang Liver cells.
