Expression of SOX9 in the Directed Differentiation of Mouse Hepatic Oval Cell into Biliary Epithelial Cells

Abstract:

ObjectiveTo investigate the expression changes of SOX9 during the directed differentiation of mouse hepatic oval cell HOCinto biliary epithelial cellBEC. MethodsA mouse HOC activation model was established by 2-acetylaminofluorene gavage and 2/3 partial hepatectomy. HOC were extracted by two-step enzyme digestion and Percoll density gradient centrifugationand then induced to differentiate into BEC by using epidermal growth factorEGF), stem cell growth factorSCFand leukemia inhibitory factorLIFin vitro. Cell morphological changes before and after differentiation were observed under inverted microscopeBEC were identified by flow cytometryand the expression of SOX9 before and after differentiation was detected by immunofluorescence.ResultsHOC were differentiated into purer BEC up to the 8th generation under the joint action of EGFSCF and LIF. SOX9 was mainly expressed in the cytoplasm of primary mouse HOCand gradually entered the nucleus during differentiationwith a significant increase in average light density value. ConclusionSOX9 enters the nucleus and its expression remarkably increases during differentiation of mouse HOCs into BECsindicating its possible involvement in the regulation of this differentiation process.
