Protective Effect of Grape Seed Proanthocyanidin Extract on Lower Extremity Arteriosclerosis Obliterans

Abstract:

Objective: To investigate the protective effect of Grape Seed Proanthocyanidin Extract(GSPE) on lower extremity
arteriosclerosis obliterans(LEASO), and to preliminarily explore its action mechanism. Methods: 30 male New Zealand rabbits were
randomly divided into three groups with 10 animals each to duplicate LEASO model. They were normal control group(CON group),
LEASO model group, and GSPE group(0.5 g/kg). The TC and ox-LDL in serum were assayed after operation for 4 th, 8 th and 12 th
week. At the end of 12th week, the content of SOD and MDA were detected. The right posterior femoral artery blood flow was detected
by color doppler ultrasound, and digital subtraction angiography(DSA)of the double posterior limbs in rabbit was conducted before
the operation and after 12th week. Results: Compared with CON group, rabbits of LEASO group had claudication and thanatosis was
found in both posterior limbs. GSPE group showed normal activity without thanatosis. Compared with LEASO group, GSPE group's
serum ox-LDL level decreased significantly at the end of 8th,12th week after operation(P<0.05). The activity of serum SOD raised
obviously while MDA decreased significantly(P<0.05). Compared with LEASO group, GSPE group showed higher PV of right
posterior superficial femoral artery under the detection of color doppler ultrasound (P<0.01). Detection of digital subtraction
angiography(DSA)showed that the distal vascular imaging of right posterior superficial femoral artery was good in GSPE group and
visible abundant collateral circulation was established. Conclusion: GSPE had a prominent protective effect on LEASO via removing
free radicals, reducing lipid peroxidation level, etc.
