Abstract:

Objective: To establish a model of isoproterenol-induced ischemic CHF in the rat, and provide a useful animal model for studies on pathogenesis and effective therapy in CHF. Methods: The model group was injected intraperitoneally(ip)with ISO at 3.0 mg/(kg/d)for 3 consecutive d and then was observed for four weeks. In the fifth week, the model group was injected (ip) sequentially with ISO at 2.0 mg/(kg·d)for 2 consecutive d and then was observed for one week. In the sixth week, the model group was injected (ip)with ISO at 3.0 mg/(kg·d)for2 consecutive d. One week later, the hemodynamic, serum cTn-Ⅰ and Ang II were measured. Lung tissue pathological changes were observed. Results: Model groups rats showed that the value of +dp/dtmax reduced 33.7 percent, and the absolute value of- dp/dtmax reduced 25.2 percent. Lung tissue pathological changes were obvious. The serum cTn- and Ang II increased significantly (P <0.01). Conclusion: Model of CHF in the rats was induced by ISO multiple and
discontinuity ip of at a certain dose and it is reliable and easy to copy and control.
