Research on the Point Mutation of K-ras Gene in Human Malignant Pleural Mesothelioma

Abstract:

Objective:To investigate K-ras gene point mutation in malignant pleural mesothelioma(MPM) tissue of patients from the crocidolite-contaminated area in Dayao County, Yunnan Province. Methods: Polymerase chain reaction(PCR)and single-strand conformation polymorphism and PCR-DNA sequencing were used to detect the point mutations of K-ras codons 12, 13 and 61 in pleural mesothelium of 59 MPM patients and 13 non-MPM patients. Results: There was no point mutation of K-ras gene in pleural mesothelial tissues of 59 MPM patients and 13 non-MPM patients, and the detection rate of K-ras gene point mutation was 0.0 %. Conclusion: The proportion of K-ras gene mutation type in MPM tissue is relatively low, which suggests that K-ras proto-oncogene point mutation does not play a key role in the induction of MPM in the crocidolite-contaminated area of Dayao county, Yunnan Province.
