Effects of the 7-Hydroxyisoflavone in the Proliferative Activity of HaCaT Cells and UVB-induced Photodamage

Abstract:

Abstract Objective To investigate the effects of the 7-Hydroxyisoflavone on the proliferative activity of HaCaT cells and the UVB-induced photodamage as well as its mechanism. Methods The cell counting assay was used to detect the effects of different concentrations of 7-Hydroxyisoflavone on the proliferative activity of HaCaT cells. Next the HaCat cells were pretreated with 200 μmol/L 7-Hydroxyisoflavone and were exposed to UVB30 mJ/cm2. Then the production of reactive oxygen speciesROS was detected by fluorescence method. The expressions of PARP1 and Caspase-8 were detected by Western blot. ResultsThe 7-Hydroxyisoflavone had the most obvious effect on the proliferative activity of HaCaT cells at the concentration of 200 μmol/L and it could reduce the production of UVB-induced ROS. Besides it could also inhibit the expressions of the PARP1 and Caspase-8.ConclusionThe 7-Hydroxyisoflavone can reduce photodamage caused by UVB and has a photoprotective effect on cells which can be used to prevent skin damage caused by UVB.
