Abstract ObjectiveTo analyze the expression level and clinical significance of thrombospondin-2 THBS2 gene mRNA in patients with malignant pleural mesothelioma MPM based on tumor databases. Methods The difference of THBS2 mRNA expression between MPM and normal tissues was analyzed by Oncomine database and the correlation between THBS2 mRNA expression and clinicopathological parameters of MPM patients was analyzed by TCGA database and UALCAN database. Kaplan- Meier survival model was constructed to analyze the prognostic effect of THBS2 gene expression on MPM patients COX regression model was constructed to explore the independent risk factors affecting the prognosis of MPM patients GEPIA2 database was used to analyze the correlation between THBS2 gene and the expression of tumor marker gene and other members of THBS gene family in MPM patients. ResultsThe expression of THBS2 gene in MPM tissues was significantly higher than that in normal pleural tissues and normal lung tissues. There were significant differences in THBS2 gene expression between N0 and N3 N1 and N3 N2 and N3 in MPM tissues. The high expression of THBS2 gene and sarcomatous and biphasic mixed tumors are independent risk factors for poor prognosis in patients with MPM. The expression of CDH11 was positively correlated with THBS2 while CALB2 and MSLN were negatively correlated with THBS2. In addition the expression of THBS1 THBS3 THBS4 was positively correlated with that of THBS2. ConclusionTHBS2 is expressed differently in different types of cancer. Through the analysis of public database it is found that the expression level of THBS2 gene is increased in human MPM tissue and the expression of THBS2 is negatively correlated with the prognosis of patients. Therefore THBS2 can be used as a potential marker for the preliminary diagnosis and prognosis of MPM.
