Abstract: Objective: To study the physiological and biochemical index and accumulation of secondary metabolites in the callus of Pueraria mirifica affected by the different levels of nitrogen( N) and kalium( K). Methods: Physiological, biochemical and accumulation of secondary metabolites in the callus of P. mirifica under different treatment conditions were studied by means of plant physiology experiment and HPLC. Results: The contents of puerarin, daidzin and genistein in the callus of P. mirifica were negatively correlated with the concentrations of NH4NO3 and KNO3 (P<0.01). The synthesis of puerarin was most favored when the concentrations of NH4NO3 and KNO3 were 1 650.0 mg/L and 0.0 mg/L, respectively. The synthesis of genistein was most favored when the concentrations of NH4NO3 and KNO3 were 825.0 mg/L and 950.0 mg/L, respectively. When the concentrations of NH4NO3 and KNO3 were 0.0 mg/L and 1 900.0 mg/L, respectively, it was most favorable for the synthesis of daidzin and total isoflavones. The content of geranylgeranyl was negatively correlated with the activities of catalase (CAT) and superoxide dismutase (SOD) (P<0.05). The content of daidzin was positively correlated with the content of malondialdehyde( MDA)( P<0.01). The content of genistein was positively correlated with MDA content and peroxidase (POD) activity (P<0.05). Conclusion: The synthesis of puerarin, daidzin and genistein, MDA content, SOD activity, POD activity and CAT activity in the callus of P. mirifica are affected by different levels of N and K.
